Hello,
We have just started doing ELISPOTS with protein instead of peptides.
Should we be allowing more time for the cells to respond to allow for
antigen processing and presentation? If so how long should we incubate
the cells for?

Yes. Insted of 20 hours at least 40 hours. There is a paper out by Carmen
Scheibenbogen's group in JIM: Schmittel A, Keilholz U, Scheibenbogen C.
