Tentagel S NH2 (90 micron mesh size beads)

density of peptide on bead is approximately 0.2-0.3 mmol/(g of Tentagel
resin)
90 micron mesh size of beads corresponds to 167 um (diameter?)

number of beads per gram is 2.86*10^6 beads

this myself. I weighed out 0.912 mg into a 1.5 mL tube, diluted to 500
uL, and then counted the beads in 10 uL fractions 3 times to get 31.3
beads/10uL -> 31.3 beads/10uL*500uL = 1567 beads in total 500 uL volume
-> 0.912mg/1567beads = 5.82*10^(-4) mg/bead or 582 ng/bead
beads

swelling volume of Tentagel S beads in water is 3.6 mL/g

Density of peptides bound to tentagel beads
  1. 25 mmol/1g*1g/3.6mL = 0.069 mmol/mL or 69 umol/mL
69 umol/dm^3*(10dm)^3/(1m)^3*1mol/1e6umol = 0.069 mol/m^3
One face of this cube would correspond to a certain density of peptide
per area
(x*mol/m)^3 =0.069*mol/m^3

density. I can determine the amount of peptide on one bead. I can then
also determine the surface area of the outside of the bead as well as
the surface area inside of the bead, and then calculate the amount of
peptide per surface area.
o How many peptides are there on one bead
- 0.25mmol/1g*1mol/1000mmol*6.02e23/1mol*1g/2.86e6beads =
5.26e13 peptides/bead

o The density of peptides will depend on the surface area.
- For example
5.26e13peptides/(0.10*4/3*Pi*(83.5)^3)*(1um)^2/(1000nm)^2 = 216
peptides/nm^2
- The larger the surface area is the lower the density of the
peptides will be.
- I need a good method for estimating the surface area of the
Tentagel beads. I e-mailed the company Rapp Polymere to see if they
have any information on this.

@ The company had no data available.
@ Zhao says that when we synthesize the beads with two
layers of lysine then the distance between the peptides will be 1
nm, which is about the same as the distance between the peptides
on the aminosilane array.
