I would like to try a few things to increase my transformation
efficiencies.
Do ethanol precipitated in-fusion products transform better with chemical
transformation or electroporation?
sample 2 and 4 compared
Does diluting the glycogen precipitated in-fusion product make a
difference?
Sample 2 and 1 compared
Does it make a difference to incubate the cells in 1 mL SOC as opposed to
about 0.1 mL SOC?
Sample 2 and 3 compared
Samples required to answer these questions.
Sample 1: ethanol precipitated in-fusion product diluted to 20 uL and 1
uL electroporated followed by 100 uL SOC incubation
Sample 2: ethanol precipitated in-fusion product diluted to 2 uL and 2 uL
electroporated followed by 100 uL SOC incubation
Sample 3: ethanol precipitated in-fusion product diluted to 2 uL and 2 uL
electroporated followed by 1000 uL SOC incubation
Sample 4: ethanol precipitated in-fusion product diluted to 2 uL and 2 uL
chemically transformed followed by 100 uL SOC incubation
