The peptides are synthesized on the tentagel resin from the C terminus

Note that on the version 2 arrays the N terminal side of the peptide is
attached to the array surface (protein sequences are typically written
from the N to C terminus). On the version 1 arrays the C terminal side of
the peptide is attached to the array surface. Selecting peptides from the
version 2 array to be attached to the Tentagel beads is not a
straightforward process. The peptide sequence cannot just be simply
reversed so that the farthest amino acids on the array are also the
farthest amino acids from the resin. When this is done, the type of link
between each amino acid will be reversed, and the peptide may not be
exactly the same. Although many antibodies and other molecules may
interact with the functional groups of the amino acids in the same way
there is no guarantee. Therefore, in order to obtain exactly the same
type of peptide on the tentagel resin as that found on the array the
peptides must be synthesized, purified, and then conjugated to the
tentagel resin.
