Infection and growth from single colony Single bacterial
colonies obtained following a procedure similar to that described for
titration above were picked and grown overnight in
2XTY-ampicillin-chloramphenicol at 30 C, 250 r.p.m. For each phage
production protocol, after overnight growth phagemids were collected from
the supernatant by centrifugation (4000 r.p.m., 30 min), and the levels
of phage production determined by titration in DH5aF cells plated on both
2XTYampicillin
and 2XTY-chloramphenicol plates.
