Gel extraction results seem to improve when DNA is extracted immediately

-DNA fragments from a screen for a slain transcript were gel extracted

immediately and the DNA sequencing results seemed to have good quality
(sequencing results from slain screen 7-28-13), whereas a potential
SMC1fs fragment was not extracted immediately and these sequencing
results were not as good (sequencing results 7-22-13).

Does it make a difference to extract DNA immediately?

Does it make a difference to mix sample by vortex vs inversion?
