++ nested pcr with tumor transcripts 9-10-13 Performed nested pcr with tumor transcripts. pcr tube layout for these pcrs and gels -"F:\kurt\storage\CIM Research Folder\DR\2013\9-10-13\gel\pcr tube layout 20130910_154247.jpg" -"F:\kurt\storage\CIM Research Folder\DR\2013\9-10-13\gel\pcr tube layout 20130910_154256.jpg" raw images of gels -F:\kurt\storage\CIM Research Folder\DR\2013\9-10-13\raw gel images\9-10-13 ^I forgot to load the ladder so I had to reload the samples -F:\kurt\storage\CIM Research Folder\DR\2013\9-11-13\raw gel images Annotated gel file here -"F:\kurt\storage\CIM Research Folder\DR\2013\9-10-13\gel\nested pcr screen gel annotated 9-10-13.svg" ^(may need to look for newer versions) see also summary spreadsheet -"F:\kurt\storage\CIM Research Folder\DR\2013\9-10-13\gel\Summary of nested pcr screen 9-10-13.xlsx" The quality of these pcr results was quite good. I probably could have done more than 30 cycles for the 1st pcr though. The R130Tr and the co1 transcripts look like they have an extra band with the tumor template which may be worth investigating. -I can get the extra band from R130_T in the 1p3 gel (10 wells from right most edge with a size around 600bp). [R130_T extra band 9-11-13] -I can get the extra band from co1_T in the 1p3 gel (3 wells from right most edge with a size around 550 bp). [co1_T extra band 9-11-13]
