In other words, does the PCR reaction have higher quality if the polymerase is not added until the reaction has already reached denaturation temperature (around 98 C)? -The answer to this question appears to be: No. This is likely because the PrimeSTAR reaction mixture is already setup for hot start performance since the mixture contains a deactivating antibody as stated in the manual. I have an experiment in which I compared hot start vs no hot start on 7-13-13 (pg. 109) of a lab notebook, and there does not appear to be any difference between the two conditions.
