Why not just do an immunoprecipitation with the purified antibody to find
the tumor antigen? Or why not flow tumor proteins past a bead with the
purified antibody on it to extract antigen.

Why not just use microscopy and do a TUNEL assay to measure killing of
cells by T cells?

does so many other things.
