azim58 - Overnight Culture of Library I plan to screen the tumor library by first making overnight cultures that combine some culture from all of the wells in each of the 41 plates in the library from the glycerol stock. Then I will have 41 pools which I can then screen for the presence of particular genes such as Shen's SMC1fs transcript and some of Hojoon's transcripts. I obtaining some culture from each well just by scraping some frozen material from each well using a 1 mL tip. This frozen material is then placed into 5 mL of LB/carb. So far I located 30 of the 41 plates all in one location (2nd from top shelf o Reese Probe 69 freezer next to -150 freezer). However, I can't find the last 11 plates. Here's a spreadsheet of the plates I have found and not found. "C:\kurt\storage\CIM Research Folder\DR\2013\4-8-13\locating plates\locating plates 4-8-13.xlsx" I ended up finding all of the plates. I had just moved them to a different shelf and I forgot about it.
