azim58 - NotI restriction digest This was a test of the NotI restriction digest to make sure that the enzyme and conditions were working appropriately. The NotI digest was for cutting the PCANTAB vector with SfiI and NotI to eventually insert scFv fragments for a phage antibody library. The NotI enzyme seemed to work just fine. An in-gel stain and a post stain were also compared. Amount of PCANTAB Cut = 800 ng Amount of Uncut PCANTAB used = 100 ng Amount of rCompTT Cut = 1000 ng Amount of uncut rCompTT = 100 ng The plasmids that were cut were cut with NotI only.
