azim58 - Cut vector DNA 060612 refer to Plan for Cloning Mouse scFv DNA 082411 Actually before I do any cloning, I need to modify the pComb3XSS vector so that it just has 1 SfiI site and 1 NotI site in the appropriate locations. In order to do this, I will cut with SpeI and NotI, and then PCR amplify this product in a manner that excludes the NotI and SfiI sites and also has sites of homology necessary for cloning the fragment back into the vector. Before modifying the vector, I just need to get more of it.
